Near as I can figure you can't. At least it's not as simple as in Win9x where you just uncheck "autoinsert notification". I looked at the help file yesterday and found that the solution is to hold down the shift key when inserting a CDROM into the drive. Kind of a weak workaround if you ask me.

There is a registry key, i can't recall it off the top of my head, but if you search MS's knowledge base you should be able to figure it out.
